Visible‐Light‐Driven Metal‐Free C−H Functionalization: Access to New Bioactive Tetrahydroisoquinoline‐Butenolide Hybrids via Domino Amine Oxidation/Vinylogous Mannich Reaction**

An efficient metal-free visible light-driven two-step domino reaction towards new bioactive tetrahydroisoquinoline-butenolide hybrid compounds was developed for the first time. Combination of fluorescein as photosensitizer and thiourea an additive found to be most effective way promote aerobic amine oxidation/vinylogous Mannich sequence with yields up 97% a broad substrate scope. Both experimental computational evidence supported crucial role singlet oxygen in C–H functionalization reaction. Furthermore, data suggest that is important due its ability act electron-transfer mediator and/or scavenger reactive species. In addition, vitro studies demonstrated their high antischistosomal anti-cancer activities.

A novel multi-component reaction for the synthesis of polyfunctionalized indoles and bis-indoles has been established. The reaction pathways were controlled by varying enamines with different substitution patterns to give polyfunctionalized indoles and bis-indoles selectively. The reaction proceeds at a fast speed within 15-30 min with water as the major byproduct, which makes work-up convenient.
